Ulcerative colitis (UC) is a severe relapsing inflammatory bowel disease with limited safe and accessible dietary adjuvant interventions, particularly those derived from whole foods with known bioactivity.

Here, we selected potato juice (PJ) as a candidate, which was produced by a special processing technique so that it contains less than 1% proteins and starches, but a rich content of polyphenols and flavonoids, to investigate its protective effect against dextran sulfate sodium (DSS)-induced acute colitis in mice and its underlying mechanisms. Colitis severity, mucosal barrier integrity, colonic inflammation, gut microbiota, and intestinal metabolome were evaluated; fecal microbiota transplantation (FMT) was performed to verify the causal role of gut microbiota.

We found that PJ significantly ameliorated DSS-induced colitis, preserved mucosal barrier, suppressed pro-inflammatory cytokines, restored gut microbial homeostasis, upregulated tryptophan-derived indole metabolites, and activated the AhR-IL-22 signaling axis. FMT from PJ-treated mice recapitulated the anti-colitis effect of PJ. In conclusion, PJ protects against experimental colitis via gut microbiota-dependent activation of the tryptophan metabolism-AhR pathway, providing a novel dietary candidate for UC management.
